Gynest (Estriol)

Gynest is prescribed for several gynecological situations where local estrogen supplementation is desired, particularly when systemic hormone therapy is not indicated or when targeted treatment of the vaginal epithelium is preferred.

Common

• Temporary vaginal irritation or burning sensation.
• Light spotting or brownish discharge.
• Mild itching or dryness.
• Slight increase in vaginal discharge volume.
• Mild breast tenderness.
• Local swelling of the vulvar area.

Serious

• Unexplained vaginal bleeding.
• Signs of estrogen excess such as nausea, headache, or fluid retention.
• Allergic reaction with rash, swelling, or difficulty breathing.
• Development of endometrial hyperplasia (rare).
• Blood clots in deep veins (rare).

Before using Gynest, certain health considerations should be reviewed to ensure safe and effective treatment.

• History of estrogen-sensitive cancers such as breast or endometrial cancer.
• Active or past thromboembolic disorders, including deep vein thrombosis or pulmonary embolism.
• Unexplained vaginal bleeding or abnormal uterine bleeding.
• Liver disease that may affect hormone metabolism.
• Pregnancy or breastfeeding status, as estriol exposure may affect the fetus or infant.
• Use in patients with known hypersensitivity to estriol or any excipients.

Estriol can interact with other medications or substances that influence estrogen pathways or clotting mechanisms.

Avoid

• Combined use with systemic estrogen therapy or high-dose hormonal contraceptives.
• Concurrent anticoagulant therapy such as warfarin without monitoring.
• Use with selective estrogen receptor modulators (SERMs) like tamoxifen.
