Herplexim (Aciclovir)

Herplexim is indicated for the treatment and management of several herpes virus infections.

• Herpes simplex virus (HSV) infections, including genital and oral lesions.
• Herpes zoster (shingles) caused by varicella‑zoster virus.
• Primary varicella (chickenpox) in immunocompetent patients.
• Recurrent herpes labialis (cold sores).
• Prevention of HSV transmission in certain clinical settings.

• Aciclovir, the active component of Herplexim, is a guanosine analogue that is selectively phosphorylated by viral thymidine kinase. The monophosphate is then converted by cellular kinases to the triphosphate form, which competitively inhibits viral DNA polymerase, halting viral DNA chain elongation and replication without significantly affecting host DNA synthesis.

Adverse reactions to Herplexim can range from mild to severe and should be monitored during therapy.

Common

• Headache
• Nausea or vomiting
• Abdominal discomfort or pain
• Fatigue or malaise
• Diarrhea

Serious

• Acute renal impairment or failure
• Severe allergic reactions such as Stevens‑Johnson syndrome
• Seizures or neurotoxicity, especially in high doses
• Blood dyscrasias including neutropenia or thrombocytopenia

• Assess renal function; dose adjustment may be required in renal impairment.

• Co‑administration with cimetidine may reduce renal clearance of aciclovir.
