• Acetylsalicylic acid irreversibly acetylates the serine residue of cyclooxygenase‑1 (COX‑1) in platelets, blocking the enzyme’s ability to convert arachidonic acid to prostaglandin H2. This inhibition prevents the formation of thromboxane A2, a potent promoter of platelet aggregation and vasoconstriction in the vascular system. Because platelets cannot synthesize new COX‑1, the antiplatelet effect persists for the lifespan of the platelet, typically 7–10 days.
