Hexyon (Haemophilus Influenzae Type B Polysaccharide (Polyribosylribitol Phosphate), Diphtheria Toxoid, Teta)

Hexyon is indicated for active immunisation against several bacterial and viral diseases in infants and young children. The vaccine provides protection by stimulating antibody production against each component antigen, reducing the risk of severe disease, hospitalization, and transmission.

• Hexyon works by presenting purified antigens from six pathogens to the immune system. The diphtheria and tetanus toxoids, pertussis proteins, hepatitis B surface antigen, inactivated poliovirus particles, and Hib polysaccharide are taken up by antigen‑presenting cells, processed, and displayed on major histocompatibility complex molecules. This activates helper T cells and B cells, leading to the production of specific IgG antibodies that neutralise the toxins, block viral entry, and promote opsonophagocytic clearance of Hib.

Local and systemic reactions may occur after administration of Hexyon. Most are mild and resolve without intervention, while rare serious events have been reported.

Common

• Pain, redness or swelling at the injection site
• Low‑grade fever (≤38.5 °C)
• Irritability or fussiness in infants
• Mild headache or fatigue

Serious

• Severe allergic reaction (anaphylaxis) with difficulty breathing
• High fever (>40 °C) or prolonged fever
• Seizures (febrile convulsions)
• Persistent crying lasting more than 3 hours
• Signs of meningitis or encephalopathy

Healthcare providers should assess each patient before giving Hexyon and follow standard vaccination precautions.

• Do not administer to individuals with a known severe allergy to any vaccine component.
• Defer vaccination in persons with a moderate or severe acute illness until recovery.
• Observe the recipient for at least 15 minutes after injection to monitor for immediate reactions.
• Use with caution in infants with a history of seizures or neurological disorders.
• Discuss the benefits and risks with pregnant or breastfeeding women, as safety data are limited.
• Record the vaccine lot number and administration site for traceability.

Hexyon may interact with other vaccines or immunomodulating agents, influencing immune response or safety.

Avoid

• Simultaneous administration with live attenuated vaccines (e.g., rotavirus) unless recommended by guidelines.
• Co‑administration with blood products that contain immunoglobulins, which could reduce vaccine efficacy.

Use with caution

• Concurrent use of immunosuppressive therapy (e.g., corticosteroids) may diminish antibody response.
• Administration with other inactivated vaccines is generally acceptable but should follow spacing recommendations.
• Patients receiving antipyretics prophylactically may experience reduced local reactogenicity, though overall immunity is not compromised.
