Humulin M3 (Mixture 3) (Insulin Human)

Humulin M3 is indicated for the management of blood glucose in patients with diabetes mellitus.

• Initiation of insulin therapy in adults with type 1 diabetes mellitus.
• Supplemental insulin for type 2 diabetes mellitus when oral agents are insufficient.
• Use in combination with basal insulin preparations to provide mealtime glucose control.
• Adjustment of dose during periods of illness, stress, or changes in diet.
• Treatment of gestational diabetes when insulin therapy is required (category B).

• Humulin M3 delivers human insulin, which binds to the insulin receptor on cell membranes. Receptor activation triggers autophosphorylation and downstream signaling through the PI3‑kinase/Akt pathway, promoting translocation of GLUT‑4 transporters to the cell surface and increasing glucose uptake in muscle and adipose tissue. Simultaneously, hepatic gluconeogenesis and glycogenolysis are suppressed, facilitating storage of glucose as glycogen and lowering circulating blood glucose levels.

Like all insulin products, Humulin M3 may be associated with adverse reactions ranging from mild to severe.

Common

• Hypoglycemia (low blood glucose) especially if meals are missed or dosing is excessive.
• Injection site reactions such as redness, swelling, or itching.
• Lipodystrophy, including lipohypertrophy at repeated injection sites.
• Weight gain associated with improved glycemic control.

Serious

• Severe hypoglycemia with neuroglycopenic symptoms (confusion, seizures, loss of consciousness).
• Allergic reactions including urticaria, angioedema, or anaphylaxis.
• Edema or fluid retention leading to peripheral swelling.
• Rare cases of insulin antibody formation causing insulin resistance.
