• Humulin NPH contains recombinant human insulin that binds to insulin receptors on muscle, adipose and hepatic cells. Receptor activation triggers the intrinsic tyrosine‑kinase pathway, promoting translocation of GLUT‑4 transporters and increasing glucose uptake. In the liver, insulin suppresses gluconeogenesis and glycogenolysis while stimulating glycogen synthesis, resulting in reduced circulating glucose concentrations over an intermediate duration. The protamine component prolongs absorption, creating a basal insulin effect lasting up to 24 hours.

Like all insulin preparations, Humulin NPH may be associated with adverse reactions, most of which are related to its glucose‑lowering action or injection site.
