• Isotretinoin binds to nuclear retinoic acid receptors, modulating gene expression that leads to decreased sebaceous gland size and reduced sebum production. It also normalizes follicular keratinization and exerts anti‑inflammatory effects, thereby limiting the proliferation of Propionibacterium acnes and reducing lesion formation.

Common Side Effects

• Dry skin and lips: increased transepidermal water loss causing xerosis and cheilitis.
• Nasal mucosa dryness: leading to epistaxis.
• Elevated liver enzymes: mild transaminase increases.
• Hypertriglyceridemia: rise in serum triglyceride levels.

Serious or Rare Side Effects

• Teratogenicity: severe birth defects if exposure occurs during pregnancy.
