Jentadueto (Metformin Hydrochloride, Linagliptin)

Jentadueto is indicated for the management of type 2 diabetes mellitus, a chronic metabolic disorder requiring glycaemic control.

• Type 2 diabetes mellitus: improves overall glycaemic control when used with diet and exercise.
• Inadequate control on metformin alone: adds DPP‑4 inhibition to further lower blood glucose.
• Combination therapy with other antidiabetics: provides complementary mechanisms to achieve target HbA1c levels.

• Metformin reduces hepatic glucose production and increases peripheral insulin sensitivity, while linagliptin inhibits the enzyme dipeptidyl peptidase‑4 (DPP‑4), leading to higher endogenous incretin levels that stimulate glucose‑dependent insulin secretion and suppress glucagon release, together producing better glycaemic regulation.

Common Side Effects

Serious or Rare Side Effects

• Lactic acidosis: rare but life‑threatening metabolic disturbance.
• Severe hypoglycaemia: especially when combined with insulin or sulfonylureas.
• Pancreatitis: inflammation of the pancreas.
• Hepatic dysfunction: abnormal liver enzyme elevations.
• Severe skin reactions: such as Stevens‑Johnson syndrome.
• Acute renal failure: sudden loss of kidney function.

Before prescribing Jentadueto, clinicians should evaluate patient‑specific risk factors and monitor relevant clinical parameters.

• Renal function: assess eGFR before initiation and periodically thereafter.
• Lactic acidosis risk: avoid in patients with severe renal or hepatic impairment.
• Hypoglycaemia: monitor closely if used with insulin or sulfonylureas.
• Pregnancy and lactation: use only if potential benefit justifies potential risk.
• Vitamin B12 deficiency: consider periodic monitoring during long‑term therapy.
