Klotrimazol Polpharma (Clotrimazole)

Clotrimazole is an azole antifungal used to treat superficial fungal infections of the skin.

• Dermatophyte infections (tinea corporis, tinea cruris, tinea pedis): Reduces fungal growth and clears the rash, itching and scaling associated with these conditions.
• Cutaneous candidiasis: Eliminates Candida species on the skin, relieving erythema, soreness and discharge.
• Seborrheic dermatitis: Controls Malassezia yeast overgrowth, improving flaking and redness of affected areas.

• Clotrimazole exerts its antifungal activity by inhibiting the fungal enzyme lanosterol 14α‑demethylase, a key component of the ergosterol biosynthesis pathway. Blocking this enzyme prevents the conversion of lanosterol to ergosterol, leading to depletion of ergosterol in the cell membrane. The resulting membrane instability disrupts fungal cell growth and replication, ultimately causing cell death. Topical application delivers the drug directly to the infected skin layers, achieving high local concentrations with minimal systemic exposure.

Clotrimazole may cause local skin reactions, most of which are mild and reversible.

Common Side Effects

• Burning sensation: Transient feeling of heat at the application site.
• Itching: Mild pruritus that usually resolves with continued use.
• Redness: Local erythema around the treated area.
• Dryness or peeling: Slight skin flaking after treatment.

Serious or Rare Side Effects

• Contact dermatitis: Allergic skin reaction with swelling, vesicles or intense itching.
• Severe skin irritation: Persistent pain, ulceration or extensive rash requiring medical attention.

• Application area: Apply only to intact skin and avoid eyes, mouth or mucous membranes.
• Allergy history: Discontinue use if signs of allergic contact dermatitis develop.
• Age restrictions: Use in children under 2 years only under medical supervision.
• Concomitant skin products: Do not apply other topical medications on the same area unless advised.
• Store at room temperature: Keep below 25 °C, in the original container, protected from moisture and direct sunlight.

Clotrimazole can interact with other topical agents and, rarely, with systemic drugs that affect skin metabolism.
