Lansoprazo Ratiopharm (Lansoprazole)

Lansoprazole is a proton pump inhibitor used to manage acid‑related gastrointestinal disorders.

• Gastro‑oesophageal reflux disease (GERD): alleviates heartburn and esophageal irritation by lowering stomach acidity.
• Peptic ulcer disease: promotes ulcer healing and prevents recurrence through acid suppression.
• Helicobacter pylori eradication (in combination therapy): enhances the effectiveness of antibiotic regimens by creating a less acidic environment.
• Zollinger‑Ellison syndrome: controls excessive gastric acid production caused by gastrin‑secreting tumors.

• Lansoprazole irreversibly binds to the H+/K+ ATPase enzyme (the gastric proton pump) located on the secretory surface of parietal cells. By inhibiting this final step of acid production, it markedly reduces basal and stimulated gastric acid secretion, leading to an increased gastric pH and allowing healing of acid‑related mucosal damage.

Lansoprazole is generally well tolerated, but adverse reactions can occur.

Common Side Effects

• Headache: mild to moderate intensity.
• Diarrhoea: usually self‑limiting.
• Nausea: occasional, may improve with food.
• Abdominal pain: mild discomfort in the stomach area.
• Flatulence: increased intestinal gas.
• Constipation: reduced bowel movements.

Serious or Rare Side Effects

• Clostridioides difficile infection: severe colitis presenting with watery diarrhoea.
• Severe allergic reactions: rash, swelling, or anaphylaxis.
• Hepatic enzyme elevation: increased ALT/AST levels.
• Hypomagnesemia: low serum magnesium with long‑term use.
• Stevens‑Johnson syndrome/toxic epidermal necrolysis: rare skin blistering disorders.
• Interstitial nephritis: inflammation of kidney tissue.

Before initiating therapy, consider the following safety measures.

• Assess renal function: adjust use in patients with severe impairment.
• Monitor for infection: be aware of increased risk of C. difficile colitis.
• Check magnesium levels: long‑term therapy may cause deficiency.
• Review concomitant medications: avoid drugs that require acidic pH for absorption.
• Store at room temperature: keep in the original pack, protect from moisture.

Lansoprazole can affect the pharmacokinetics of several medicines.

Major Interactions (Avoid)

• Ketoconazole: reduced absorption and efficacy.
• Atazanavir: decreased plasma concentrations.
• Clopidogrel: impaired activation leading to reduced antiplatelet effect.

Moderate Interactions (Monitor Closely)

• Warfarin: possible increase in INR, monitor coagulation.
• Phenytoin: may lower phenytoin levels, adjust dose if needed.
• Digoxin: potential rise in serum digoxin, observe for toxicity.
