Tioguanine is converted inside cells to thioguanine nucleotides, which block purine synthesis and become incorporated into DNA and RNA. This disrupts nucleic‑acid function, leading to cytotoxicity of rapidly dividing cells and suppression of immune‑cell proliferation.
