Lanzol (Lansoprazole)

Lansoprazole is a proton pump inhibitor used to reduce gastric acid production in several acid‑related conditions.

• Gastroesophageal reflux disease: alleviates heartburn and promotes healing of esophageal irritation.
• Erosive esophagitis: reduces acid exposure to allow mucosal recovery.
• Peptic ulcer disease: supports ulcer healing and prevents recurrence.
• Helicobacter pylori eradication therapy: enhances the effectiveness of antibiotic regimens by suppressing gastric acid.

• Lansoprazole selectively and irreversibly binds to the H+/K+ ATPase enzyme (the gastric proton pump) located on the secretory surface of parietal cells. By inhibiting this final step of acid secretion, it markedly reduces both basal and stimulated gastric acid output, leading to increased gastric pH and providing an environment conducive to mucosal healing. The inhibition persists for the lifespan of the pump, typically 24‑48 hours, which allows once‑daily dosing in most patients.

Lansoprazole may cause a range of adverse reactions, most of which are mild and reversible.

Common Side Effects

• Headache: mild to moderate pain often resolving without treatment.
• Diarrhea: loose stools that may improve with continued therapy.
• Nausea: feeling of stomach upset, usually transient.
• Abdominal pain: discomfort in the stomach area.

Serious or Rare Side Effects

• Clostridioides difficile infection: severe colitis requiring medical attention.
• Severe allergic reaction (anaphylaxis): rapid onset of rash, swelling, and breathing difficulty.
• Low magnesium levels (hypomagnesemia): can lead to cardiac arrhythmias and muscle weakness.

When prescribing Lanzol, clinicians should consider several safety measures.

• Assess renal function: dose adjustment may be needed in severe renal impairment.
• Monitor magnesium: prolonged therapy can cause hypomagnesemia.
• Avoid abrupt discontinuation: may lead to rebound acid hypersecretion.
• Use with caution in patients with liver disease: hepatic metabolism may be affected.
• Store at room temperature: keep in the original container, protect from moisture and light.

Lansoprazole can affect the absorption or metabolism of several co‑administered drugs.

Major Interactions (Avoid)

• Atazanavir: reduced absorption of atazanavir leading to therapeutic failure.
• Clopidogrel: decreased activation of clopidogrel, reducing antiplatelet effect.
• Rilpivirine: lowered plasma concentrations, compromising HIV therapy.

Moderate Interactions (Monitor Closely)

• Warfarin: possible increase in INR, monitor coagulation parameters.
• Methotrexate: reduced renal clearance, monitor serum levels.
• Digoxin: increased serum digoxin concentrations, monitor for toxicity.
