How It Works

  • Lapatinib binds to the intracellular ATP‑binding site of the HER2 (ErbB2) and epidermal growth factor receptor (EGFR) tyrosine kinases, preventing autophosphorylation and downstream activation of the PI3K/AKT and MAPK pathways. By blocking these signaling cascades, the drug reduces tumor cell proliferation, survival and angiogenesis. Additionally, inhibition of HER2‑driven heterodimer formation limits cross‑talk with other ErbB family members, further suppressing oncogenic signaling.
