Lenalidomide Grindeks (Lenalidomide)

Lenalidomide is used in the management of hematologic cancers and related blood disorders.

• Lenalidomide binds to cereblon, a component of the CRL4^CRBN E3 ubiquitin ligase complex, altering its substrate specificity. This triggers ubiquitination and proteasomal degradation of transcription factors such as Ikaros and Aiolos, leading to modulation of cytokine production, inhibition of angiogenesis, and direct anti‑tumor activity against malignant plasma and lymphoid cells.

Lenalidomide may cause a range of adverse reactions, varying from mild to severe.

Common Side Effects

• Neutropenia: reduced neutrophil count.
• Thrombocytopenia: low platelet levels.
• Anemia: decreased red blood cells.
• Fatigue: persistent tiredness.
• Nausea: upset stomach and vomiting.
• Diarrhea: loose stools.

Serious or Rare Side Effects

• Venous thromboembolism: deep vein thrombosis or pulmonary embolism.
• Severe neutropenia: heightened infection risk.
• Severe thrombocytopenia: increased bleeding risk.
• Secondary primary malignancies: emergence of new cancers.
• Severe skin reactions: Stevens‑Johnson syndrome or toxic epidermal necrolysis.
