Indications & Clinical Uses

Linezolid is indicated for bacterial infections affecting the skin, lungs and resistant Gram‑positive organisms.

  • Complicated skin and soft‑tissue infections: effective against MRSA and other Gram‑positive pathogens, helping resolve infection and promote healing.
  • Community‑acquired and nosocomial pneumonia: achieves bacterial clearance in lung tissue, improving respiratory function.
  • Infections caused by methicillin‑resistant Staphylococcus aureus (MRSA): provides targeted activity where other agents may fail.
  • Infections caused by vancomycin‑resistant Enterococcus (VRE): offers an alternative treatment option for resistant enterococcal infections.

How It Works

  • Linezolid inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit. This interaction prevents formation of the initiation complex, blocking translation of essential proteins and halting bacterial growth. The binding is reversible and selective for bacterial ribosomes, resulting in a bacteriostatic effect against most susceptible Gram‑positive organisms. It does not significantly affect human mitochondrial ribosomes at therapeutic concentrations, contributing to its safety profile.

Side Effects

Linezolid may cause a range of adverse reactions, varying from mild to severe.

Common Side Effects

  • Nausea: mild gastrointestinal upset occurring in many patients.
  • Diarrhea: loose stools that may be self‑limiting.
  • Headache: transient cranial discomfort.
  • Vomiting: occasional emesis.
  • Metallic taste: altered taste perception.
