Nailex (Amorolfine Hydrochloride)

• Nailex is used to manage fungal nail infections by targeting the causative organisms and limiting disease progression.
• Onychomycosis: reduces fungal colonisation of toenails and fingernails, improving nail appearance and preventing spread.
• Fungal nail infection: alleviates symptoms such as discoloration, thickening and brittleness of the nail plate.
• Prevention of recurrence: maintains nail health after successful treatment, lowering the chance of reinfection.

• Amorolfine inhibits ergosterol synthesis by blocking the fungal enzymes Δ14‑reductase and Δ7‑Δ8‑isomerase. This disruption prevents formation of a functional cell membrane, leading to loss of membrane integrity and ultimately fungal cell death, while sparing human cells that do not rely on ergosterol.

Local application of Nailex may cause mild to moderate reactions at the site of use.

Common Side Effects

• Burning sensation: transient feeling of heat at the application site.
• Itching: mild pruritus that usually resolves with continued use.
• Redness: localized erythema around the nail.
• Dryness: skin around the nail may become dry or flaky.
• Scaling: slight flaking of the surrounding skin.

Serious or Rare Side Effects

• Allergic contact dermatitis: severe rash, swelling, or blistering requiring medical attention.
• Severe skin irritation: persistent pain, ulceration, or necrosis.
• Systemic absorption reactions: rare systemic effects such as liver enzyme elevation.
• Anaphylaxis: extremely rare, rapid onset of breathing difficulty and swelling.

• Avoid use on broken or inflamed skin: apply only to intact nail and surrounding skin.
• Do not apply to mucous membranes: keep product away from eyes, mouth and genital areas.
• Discontinue if severe irritation occurs: stop treatment and seek medical advice.
