Indications & Clinical Uses

Omeprazole is a proton pump inhibitor that acts on the gastric acid secretion pathway. It is indicated for several acid‑related disorders:

  • Gastro‑oesophageal reflux disease (GERD): Improves symptom relief and promotes healing of the esophageal lining.
  • Peptic ulcer disease: Facilitates ulcer healing and reduces recurrence risk.
  • Zollinger‑Ellison syndrome: Controls excess gastric acid production.
  • Erosive esophagitis: Aids mucosal healing and symptom control.
  • NSAID‑induced ulcer prophylaxis: Lowers risk of ulcer development in at‑risk patients.

How It Works

  • Omeprazole selectively inhibits the H+/K+‑ATPase enzyme (the gastric proton pump) located on the secretory surface of gastric parietal cells. By binding covalently to the pump’s cysteine residues, it blocks the final step of acid secretion, leading to a prolonged reduction in gastric acidity and an increase in gastric pH.

Side Effects

Adverse reactions may range from mild to severe and should be monitored during therapy.

Common Side Effects

  • Headache: mild, transient headache.
  • Diarrhea: loose stools, often self‑limiting.

  • Review hepatic status: use cautiously in liver disease.
  • Evaluate infection risk: monitor for Clostridioides difficile if prolonged therapy is planned.
  • Pregnancy and lactation: weigh benefits against potential risks; consult obstetric guidance.
  • Drug‑interaction review: check concomitant medications for possible interactions.
  • Store at ambient temperature: keep below 25°C, retain original packaging, protect from moisture.
