Nyxoid (Naloxone)

Naloxone is an opioid antagonist used in the management of opioid‑related emergencies.

• Opioid overdose: quickly restores normal breathing in individuals with life‑threatening respiratory depression.
• Opioid‑induced respiratory depression: reverses breathing suppression caused by opioid analgesics.
• Post‑opioid administration: provides a safety net after procedures involving opioid pain control.

• Naloxone competitively binds to mu‑opioid receptors in the central nervous system, displacing opioid molecules and blocking further receptor activation. This rapid antagonism restores normal neuronal signaling and reverses the respiratory and CNS depression caused by opioid agonists.

Adverse reactions to nasal naloxone are generally mild and transient.

Common Side Effects

• Nasal irritation: mild burning or stinging sensation at the site of administration.
• Headache: mild to moderate headache following use.
• Dizziness: transient light‑headedness after the dose.
• Nausea: occasional feeling of nausea.
• Sweating: increased perspiration.
• Cough: brief cough reflex triggered by spray.

Serious or Rare Side Effects

• Acute withdrawal: severe opioid withdrawal symptoms in dependent individuals.
• Hypertension: sudden increase in blood pressure.
• Cardiac arrhythmia: irregular heart rhythm.
• Seizure: rare seizure activity.
• Anaphylaxis: life‑threatening allergic reaction.
• Pulmonary edema: fluid accumulation in the lungs.

Naloxone should be used only in settings where medical supervision is available.

• Assess patient: confirm opioid overdose before administration.
• Monitor vitals: observe breathing, heart rate and consciousness after use.
• Re‑dose if needed: a second dose may be required if respiratory depression persists.
• Avoid in hypersensitivity: do not use if the patient has known allergy to naloxone or formulation components.
• Store at ambient temperature: keep in original packaging, protect from moisture and direct sunlight.
