  • Colorectal cancer: improves survival when combined with fluoropyrimidine‑based regimens.
  • Gastric cancer: used in combination protocols for advanced disease.
  • Pancreatic cancer: part of multi‑drug regimens in selected cases.
  • Metastatic solid tumours: incorporated into clinical trial‑based combination therapies.
  • Peri‑operative treatment of colorectal tumours with curative intent (Adjuvant therapy post‑surgery).

How It Works

  • Oxaliplatin exerts its antitumour activity by forming platinum‑DNA adducts that distort the DNA helix, inhibiting replication and transcription. The resulting DNA cross‑links trigger cell‑cycle arrest and apoptosis, particularly in rapidly dividing cancer cells. Its activity is enhanced when combined with fluoropyrimidines, which interfere with DNA synthesis pathways.

Side Effects

Oxaliplatin may cause a range of adverse reactions, most of which are dose‑dependent and reversible.

Common Side Effects

  • Nausea and vomiting: frequent during or shortly after infusion.
  • Peripheral neuropathy: tingling or numbness, often cumulative.
  • Diarrhoea: may range from mild to moderate.
  • Fatigue: generalised tiredness during treatment cycles.
  • Thrombocytopenia: reduced platelet count observed in blood tests.

Serious or Rare Side Effects

  • Severe hypersensitivity reactions: anaphylaxis or bronchospasm.
  • Acute neurotoxicity: sudden onset of muscle cramps or jaw tightness.
  • Hemolytic‑uremic syndrome: rare renal‑related complication.
  • Severe neutropenia: marked drop in neutrophil count increasing infection risk.
  • Cardiotoxicity: uncommon arrhythmias or myocardial injury.
