Indications & Clinical Uses

Omeprazole is a proton pump inhibitor that acts on the gastric acid secretion pathway. It is indicated for several acid‑related conditions:

  • Gastro‑oesophageal reflux disease (GERD): alleviates heartburn and promotes healing of the oesophageal lining.
  • Peptic ulcer disease: supports ulcer healing and reduces risk of recurrence.
  • Zollinger‑Ellison syndrome: controls excessive gastric acid production.
  • Erosive oesophagitis: heals erosive damage to the oesophageal mucosa.
  • NSAID‑induced ulcer prophylaxis: lowers ulcer risk in patients requiring long‑term NSAID therapy.

How It Works

  • Omeprazole irreversibly binds to the H+/K+‑ATPase enzyme (the gastric proton pump) located on the secretory surface of parietal cells in the stomach. By inhibiting this pump, it blocks the final step of acid secretion, leading to a sustained reduction in gastric acidity and an increase in gastric pH.

Side Effects

Omeprazole is generally well tolerated, but adverse reactions can occur.

Common Side Effects

  • Headache: mild to moderate intensity.
  • Diarrhea: usually transient.
  • Abdominal pain: occasional cramping.
  • Nausea: mild and self‑limiting.
  • Flatulence: mild bloating.

Serious or Rare Side Effects

  • Clostridioides difficile infection: severe colitis risk.
  • Acute interstitial nephritis: renal inflammation.
  • Hypomagnesemia: low serum magnesium levels.
  • Stevens‑Johnson syndrome: severe skin reaction.
  • Fracture risk: possible reduction in bone mineral density.
  • Vitamin B12 deficiency: impaired absorption with long‑term use.
