Indications & Clinical Uses

Palonosetron is a selective 5‑HT3 receptor antagonist used in the management of nausea and vomiting related to specific therapeutic pathways.

  • Chemotherapy‑induced nausea and vomiting (CINV): Provides effective prophylaxis for acute and delayed phases in adult patients receiving moderately to highly emetogenic chemotherapy.
  • Post‑operative nausea and vomiting (PONV): Reduces the incidence of nausea and vomiting after surgical procedures requiring general anesthesia.
  • Radiotherapy‑induced nausea and vomiting: Helps mitigate the nausea associated with radiation treatment of the abdomen or pelvis.
  • Antiemetic prophylaxis in patients receiving highly intense multimodal anti‑emetic regimens.
  • Adjunctive therapy for breakthrough nausea in patients already on a standard anti‑emetic protocol.
  • Support for patients under palliative care with refractory nausea that is not controlled by first‑line agents.

How It Works

  • Palonosetron binds with high affinity to serotonin type 3 (5‑HT3) receptors located on vagal afferent nerves in the gastrointestinal tract and the central nervous system. This binding blocks serotonin‑mediated signaling that triggers the vomiting reflex, providing a prolonged anti‑emetic effect that lasts up to 72 hours after a single dose.
