  • Propranolol hydrochloride is a stable, white, crystalline solid which is readily soluble in water and ethanol. Its molecular weight is 295.80.
  • Propranolol hydrochloride is available as 10u00a0mg, 20u00a0mg, 40u00a0mg, 60u00a0mg, and 80u00a0mg tablets for oral administration.
  • The inactive ingredients contained in Propranolol Hydrochloride Tablets USP are: anhydrous lactose, colloidal silicon dioxide, croscarmellose sodium, D&C Yellow #10 (10 mg, 40 mg and 80 mg tablets), FD&C Blue #1 (20 mg tablet), FD&C Blue #2 (40 mg tablet), FD&C Red #40 (60 mg tablet), FD&C Yellow #6 (10 mg and 80 mg tablets), magnesium stearate, and microcrystalline cellulose.

  • Propranolol is contraindicated in 1) cardiogenic shock; 2) sinus bradycardia and greater than first degree block; 3) bronchial asthma; and 4) in patients with known hypersensitivity to propranolol hydrochloride.

  • Propranolol is not significantly dialyzable. In thenevent of overdosage or exaggerated response, the following measuresnshould be employed:
  • General: If ingestion isnor may have been recent, evacuate gastric contents, taking care tonprevent pulmonary aspiration.
  • Supportive Therapy:nHypotension and bradycardia have been reported following propranololnoverdose and should be treated appropriately. Glucagon can exert potentninotropic and chronotropic effects and may be particularly usefulnfor the treatment of hypotension or depressed myocardial functionnafter a propranolol overdose. Glucagon should be administered as 50-150u00a0mcg/kgnintravenously followed by continuous drip of 1-5 mg/hour for positivenchronotropic effect. Isoproterenol, dopamine or phosphodiesteraseninhibitors may also be useful. Epinephrine, however, may provoke uncontrollednhypertension. Bradycardia can be treated with atropine or isoproterenol.nSerious bradycardia may require temporary cardiac pacing.
  • The electrocardiogram, pulse, blood pressure, neurobehavioralnstatus and intake and output balance must be monitored. Isoproterenolnand aminophylline may be used for bronchospasm.
